The bacterial community composition and diversity in rock varnish of Turpan Basin were investigated by restriction fragment length polymorphism (RFLP) and clone library of the 16S rRNA gene. 114 positive clones were screened, which could be grouped into 28 phylotypes and then further divided into 23 different operational taxonomic units (OTUs). These were affiliated into 5 phyla (Acidobacteria, Proteobacteria, Chloroflexi, Firmicutes and Cyanobacteria). Clones from actinobacteria were the dominant, accounting for 67.5% of total clones in the library, followed by Proteobacteria (15.8%), Chloroflexi (13.2%), Firmicutes (2.6%) and Cyanobacteria (0.9%). Rubrobacter (accounts for 35%) in the phylum Actinobacteria was the dominant genus and contained many species which might be resistant to gamma radiation. A 70% of the library clone sequences showed less 97% similarity to 16S rRNA gene sequences of standard strains obtained by pure culture. Shannon–Wiener index value of this study is 2.52 and is lower than deep-sea sediments, soils, lakes and other environments. Results of this study showed that bacterial diversity in rock varnishes of Turpan Basin was low, but maybe exist a large number of new unknown taxons, especially species that could well adapted to drought and resist radiation.  相似文献

Summary With increasing frequency, epidemiologic studies are addressing hypotheses regarding gene‐environment interaction. In many well‐studied candidate genes and for standard dietary and behavioral epidemiologic exposures, there is often substantial prior information available that may be used to analyze current data as well as for designing a new study. In this article, first, we propose a proper full Bayesian approach for analyzing studies of gene–environment interaction. The Bayesian approach provides a natural way to incorporate uncertainties around the assumption of gene–environment independence, often used in such an analysis. We then consider Bayesian sample size determination criteria for both estimation and hypothesis testing regarding the multiplicative gene–environment interaction parameter. We illustrate our proposed methods using data from a large ongoing case–control study of colorectal cancer investigating the interaction of N‐acetyl transferase type 2 (NAT2) with smoking and red meat consumption. We use the existing data to elicit a design prior and show how to use this information in allocating cases and controls in planning a future study that investigates the same interaction parameters. The Bayesian design and analysis strategies are compared with their corresponding frequentist counterparts.  相似文献

For most ecosystems, net ecosystem exchange of CO2 (NEE) varies within and among years in response to environmental change. We analyzed measurements of CO2 exchange from eight native rangeland ecosystems in the western United States (58 site‐years of data) in order to determine the contributions of photosynthetic and respiratory (physiological) components of CO2 exchange to environmentally caused variation in NEE. Rangelands included Great Plains grasslands, desert shrubland, desert grasslands, and sagebrush steppe. We predicted that (1) week‐to‐week change in NEE and among‐year variation in the response of NEE to temperature, net radiation, and other environmental drivers would be better explained by change in maximum rates of ecosystem photosynthesis (Amax) than by change in apparent light‐use efficiency (α) or ecosystem respiration at 10 °C (R10) and (2) among‐year variation in the responses of NEE, Amax, and α to environmental drivers would be explained by changes in leaf area index (LAI). As predicted, NEE was better correlated with Amax than α or R10 for six of the eight rangelands. Week‐to‐week variation in NEE and physiological parameters correlated mainly with time‐lagged indices of precipitation and water‐related environmental variables, like potential evapotranspiration, for desert sites and with net radiation and temperature for Great Plains grasslands. For most rangelands, the response of NEE to a given change in temperature, net radiation, or evaporative demand differed among years because the response of photosynthetic parameters (Amax, α) to environmental drivers differed among years. Differences in photosynthetic responses were not explained by variation in LAI alone. A better understanding of controls on canopy photosynthesis will be required to predict variation in NEE of rangeland ecosystems.  相似文献

Droughts are expected to become more frequent under global climate change. Avifauna depend on precipitation for hydration, cover, and food. While there are indications that avian communities respond negatively to drought, little is known about the response of birds with differing functional and behavioural traits, what time periods and indicators of drought are most relevant, or how response varies geographically at broad spatial scales. Our goals were thus to determine (1) how avian abundance and species richness are related to drought, (2) whether community variations are more related to vegetation vigour or precipitation deviations and at what time periods relationships were strongest, (3) how response varies among avian guilds, and (4) how response varies among ecoregions with different precipitation regimes. Using mixed effect models and 1989–2005 North American Breeding Bird Survey data over the central United States, we examined the response to 10 precipitation‐ and greenness‐based metrics by abundance and species richness of the avian community overall, and of four behavioural guilds. Drought was associated with the most negative impacts on avifauna in the semiarid Great Plains, while positive responses were observed in montane areas. Our models predict that in the plains, Neotropical migrants respond the most negatively to extreme drought, decreasing by 13.2% and 6.0% in abundance and richness, while permanent resident abundance and richness increase by 11.5% and 3.6%, respectively in montane areas. In most cases, response of abundance was greater than richness and models based on precipitation metrics spanning 32‐week time periods were more supported than those covering shorter time periods and those based on greenness. While drought is but one of myriad environmental variations birds encounter, our results indicate that drought is capable of imposing sizable shifts in abundance, richness, and composition on avian communities, an important implication of a more climatically variable future.  相似文献

A simple offline LC–MS/MS method for the quantification of sitagliptin in human plasma is described. Samples are prepared using protein precipitation. Filtration of the supernatants through a Hybrid-SPE-PPT plate was found to be necessary to reduce ionization suppression caused by co-elution of phospholipids with sitagliptin. The sitagliptin and its stable isotope labeled internal standard (IS) were chromatographed under hydrophilic interaction chromatography conditions on a Waters Atlantis HILIC Silica column (2.1 mm × 50 mm, 3 μm) using a mobile phase of ACN/H2O (80/20, v/v) containing 10 mM NH4Ac (pH 4.7). The sample drying after protein precipitation due to high organic content in the sample is not necessary, because HILIC column was used. The analytes were detected with a tandem mass spectrometer employing a turbo ion spray (TIS) interface in positive ionization mode. The multiple reaction monitoring (MRM) transitions were m/z 408 → 235 for sitagliptin and m/z 412 → 239 for IS. The lower limit of quantitation (LLOQ) for this method is 1 ng/mL when 100 μL of plasma is processed. The linear calibration range is 1–1000 ng/mL for sitagliptin. Intra-day precision and accuracy were assessed based on the analysis of six sets of calibration standards prepared in six lots of human control plasma. Intra-day precision (RSD%, n = 6) ranged from 1.2% to 6.1% and the intra-day accuracy ranged from 97.6% to 103% of nominal values.  相似文献

We study the spread of susceptible-infected-recovered (SIR) infectious diseases where an individual's infectiousness and probability of recovery depend on his/her “age” of infection. We focus first on early outbreak stages when stochastic effects dominate and show that epidemics tend to happen faster than deterministic calculations predict. If an outbreak is sufficiently large, stochastic effects are negligible and we modify the standard ordinary differential equation (ODE) model to accommodate age-of-infection effects. We avoid the use of partial differential equations which typically appear in related models. We introduce a “memoryless” ODE system which approximates the true solutions. Finally, we analyze the transition from the stochastic to the deterministic phase.  相似文献

The frequency of mating in insects is often an important determinant of female reproductive output and male sperm competition. In Lepidoptera that provide male nutrients to the female when mating, it is hypothesized that polyandry may be more prevalent. This is thought to be especially so among species described as income breeders; that is, in species who do not derive all their nutrients for reproductive output entirely from the resources obtained during the larval stage. We selected the geometrid moth, Mnesampela privata (Guenée) (Lepidoptera: Geometridae), to examine this hypothesis further. We found this species was best characterized as an income breeder with female weight on emergence positively correlated with total egg load but not with the number of eggs laid. Further, in accord with income breeders, females emerged with a partially developed egg load and lifetime fecundity was positively correlated with the number of oviposition days. However, in the laboratory we found that incidence of repeated matings or polyandry was rare. When moths were paired singly over their lifetime, only 4% of mated females multiple mated. When females were paired with three males concurrently, female mating success increased from 60 to 81% with multiple mating among mated females increasing to just 15%. Dissection of wild caught M. privata found that polyandry levels were also low with a maximum of 16.4% of females collected at any one time being multiple mated. In accord with theory, mating significantly increased the longevity of females, but not of males, suggesting that females acquire essential resources from male ejaculates. Despite this, multiple mated females showed a trend toward decreasing rather than increasing female reproductive output. Spermatophore size, measured on death of the female, was not correlated with male or female forewing length but was negatively correlated with the number of fertile eggs laid and female longevity. Smaller spermatophore width may be related to uptake of more nutrients by the female from a spermatophore. We discuss our findings in relation to income breeding and its relationship to polyandry in Lepidoptera.  相似文献

Many long‐lived plant and animal species have nondiscrete overlapping generations. Although numerous models have been developed to predict the effective sizes (Ne) of populations with overlapping generations, they are extremely difficult to apply to natural populations because of the large array of unknown and elusive life‐table parameters involved. Unfortunately, little work has been done to estimate the Ne of populations with overlapping generations from marker data, in sharp contrast to the situation of populations with discrete generations for which quite a few estimators are available. In this study, we propose an estimator (EPA, estimator by parentage assignments) of the current Ne of populations with overlapping generations, using the sex, age, and multilocus genotype information of a single sample of individuals taken at random from the population. Simulations show that EPA provides unbiased and accurate estimates of Ne under realistic sampling and genotyping effort. Additionally, it yields estimates of other interesting parameters such as generation interval, the variances and covariances of lifetime family size, effective number of breeders of each age class, and life‐table variables. Data from wild populations of baboons and hihi (stitchbird) were analyzed by EPA to demonstrate the use of the estimator in practical sampling and genotyping situations.  相似文献

2009年9~12月以4只大猩猩(Gorilla gorilla)为研究对象,应用瞬时扫描取样法观察记录其活动行为,建立不同取样间隔与数据准确性和数据独立性的线性回归方程,计算变量系数和常数项的95%置信区间,进而确定瞬时扫描法的最适取样间隔。研究结果表明,大猩猩活动主要集中于上午10:00~11:00时和下午14:00~15:00时(χ2=19.110,df=4,P=0.001),不同大猩猩个体间活动行为存在显著差异(Kruskal Wallis Test,χ2=11.229,df=3,P=0.011)。随着取样间隔的增大,间隔取样与连续取样数据的相关系数呈下降趋势,满足数据准确性需求的最适取样间隔为3~13 min,随取样间隔的增大,数据独立可能性逐渐升高,其中满足数据独立性需求的最适取样间隔为2~8 min。综合考虑以上两种因素,确定针对大猩猩活动行为的瞬时扫描取样的最适间隔为3~8 min。  相似文献

季节性干旱对湖南省农业生产造成很大影响,明确防旱避灾种植制度,可为指导当地农业生产提供科学依据。基于湖南省代表站点1981-2007年的气候数据和作物生育期资料,依据干燥度和地形地势特点,划分不同类型区,并在各区域内选取代表站点和代表性种植制度,采用FAO推荐的分段单值平均作物系数法订正了作物系数。比较了主要种植制度的作物需水与自然降水适配度及作物需水与自然降水适配度保证指数,确定了湖南省基于自然降水的防旱避灾种植制度。研究结果表明:各区域基于自然降水资源的防旱避灾种植制度分别为:(1)湘中偏北湿润中低海拔地形复杂区和湘东北半湿润中低海拔山地丘陵区为麦-稻和薯-稻种植模式;(2)湘北半湿润低海拔平原区为油-棉、油-稻和豆-稻种植模式;(3)湘东偏北半干旱低海拔平原与盆地丘陵过渡区为油-苕、油-棉和油-稻种植模式;(4)湘东南较干旱中低海拔丘陵区为油-稻、麦-稻和薯-稻种植模式;(5)中南部较干旱中低海拔盆地区为油-稻和油-棉种植模式;(6)湘西北较湿润中高海拔中山低山区为薯-稻和油-苕种植模式;(7)湘西南较湿润中高海拔丘陵区为油-苕、油-稻、麦-稻和薯-稻种植模式;(8)湘西干旱高海拔山地丘陵区为油-苕和油-棉种植模式。  相似文献
